With the increasing number of vendors selling library construction kits and reagents library construction may seem to be a fairly straightforward step in the sequencing process. And for many researchers it can be for most libraries once you have sample QC steps in place and completely understand the process. There are however exceptions. One of which are mate pair libraries. There has been increasing evidence on the difficulty level and complexity in preparation of these libraries. If you’ve tried to build a mate pair library several times and sworn them off entirely, you are not alone.

At Cofactor Genomics we’ve put together a mate pair library protocol with an 84% first run success rate. We are committed to making mate pair libraries work because we believe they add significant value to an assembly. Our custom modified protocol generates mate pairs that can overcome the usual hurdles like running into the adapter. We have also set sample QC in place, because good starting material is crucial in generating a successful and well diverse mate pair library.
